IL-29, also termed IFN λ1, belongs to the IFN type III family. IL-29 is a 20 kDa secreted monomeric protein.

Interleukin 29 plays a role in antiviral defence, predominantly in the epithelial tissues. It has been described as a therapeutic agent to treat patients with chronic hepatitis C virus by inhibiting B and C hepatitis virus replication. It also inhibits the Th2 response and acts directly on CD4+ T-Cells and dendritic cells.

Its anti-proliferative and anti-tumour activities make it a novel target for the treatment and control of some cancers.

 

Interleukin 34 (IL-34) plays a role in macrophage differentiation and proliferation. The IL-34 protein is composed of 241 amino acids, with a molecular mass of 39 kDa, secreted as homodimer. It is expressed in various tissues and it is most abundant in the spleen.

Similar to M-CSF, IL-34 plays a major role in osteoclast genesis: in the presence of RANKL, IL-34 induces the proliferation and adhesion of osteoclast progenitors in vitro.

IL-34 has been implicated in some disease processes such as rheumatoid arthritis (RA). Joint fluid levels of IL-34 are increased in RA patients compared to controls with osteoarthritis.
